  CHORI-1106 Brugia malayi Fosmid Library   

The CHORI-1106 (fosmid) library has been constructed by Chung-Li Shu and Kazutoyo Osoegawa in Pieter de Jong's laboratory, BACPAC Resources, Children's Hospital Oakland Research Institute. Agarose-embedded genomic DNA was prepared from Brugia malayi (TRS strain) by Dr. Jeremy Foster (New England Boilabs). To create fosmid clones, genomic DNA was sheared by passing through a Hamilton syringe. The sheared fragments were size-purified using pulsed-field gel electrophoresis then end-polished by T4 DNA polymerase treatment. The end-repaired DNA was ligated with pEpiFOS5 vector (Epicenter) and the ligation products were incubated with "Gigapack III Gold" (Stratagene) packaging extracts. The lambda-packaged DNA was transfected into the DH5?MCR strain. The CHORI-1106 library is comprised of 9,000 clones (plates 25-48) with an average insert size of 43 kb. The total of 9,000 clones from the library provides ~3.5X haploid genome coverage. The arrayed clones have also been gridded together with CHORI-106 BAC library onto a 22x22cm nylon high-density filter for screening by probe hybridization. Each hybridization membrane represents about 9,000 distinct BAC and 9,000 fosmid clones, each represented in duplicate. Library construction was supported by a sub-contract from a grant (NIH/NIAID U01-AI50903-02) awarded to Dr. Elodie Ghedin at The Institute for Genomic Research (TIGR).

Data on the CHORI-1106 clone average insert size has been determined by Pulsed Field Gel Electrophoresis.

The library is available as library plates, individual clones, and high-density hybridization sets (see Filter Availability). The library is available for academic users by reimbursement of duplication costs (labor & materials), resulting in a price of $20 per 384-well dish. Dishes can only be obtained per BAC and fosmid libraries (CHORI-106 and CHORI-1106: $960) or per fosmid library ($480).Individual clones identified after screening can be obtained as stab cultures at a cost of $25 per clone in addition to a $35 set-up charge per shipment. Shipping will be done by Federal Express and charged to the recipients account number.
